Procedure
A. Sample Preparation
1. BLL Solutions in Chloroform:Methanol (2:1)
a. If the sample is not a clear solution, warm it in a 37°C water bath for about 5-10 minutes. Shake it vigorously then let it cool to room temperature. If the sample is a clear solution, just bring it to room temperature. When all samples reach room temperature, shake vigorously and transfer duplicate 2.0 mL aliquots to suitable test tubes.
b. Evaporate the 2-mL aliquots to dryness under a stream of nitrogen or reduced pressure at 40°C. Add 3 mL of chloroform:methano1 (81:15) to dissolve the residue and re-evaporate it to complete dryness. Repeat this step as necessary to ensure dryness of the residue (Note 1).
c. Add 4 mL of hexane to each evaporated sample and slurry the residue by sonicating for about 30 seconds. In case there is some residue still left, continue sonicating in bursts of five seconds until the residue is well suspended in hexane.
2. Surfactant TA, Lyophi1ized Powder (Prepare in duplicate)
Weigh accurately about 35 mg of surfactant ТА. Add 4 mL of hexane. Sonicate for 30 seconds.
3. Surfactant TA, Aqueous Suspension
a. Using a “to-contain” pipet, dilute 5 ml of the sample to 100 mL with CHCl3:MeOH (2:1). The sample should be thawed and gently mixed before taking the 5 mL aliquot.
b. For each sample, evaporate duplicate 20-mL aliquots under N2 and at 45°C, to approximately 10 mL, then add 10 mL CHCl3. Repeat the above step and evaporate to dryness.
c. Wash the sides down with 5 ml CHCl3:MeOH (81:15) and evaporate to dryness. Repeat using only 3 mL CHCl3 : MeOH (81:15).
d. Add 4 mL of hexane, sonicate for about 30 seconds or until the residue is well suspended.

Что это означает? Что промывается Промывают сторонами вниз? Какими сторонами .. или просто боковые поверхности?? Сапсем запутался ....

И аналогично:
D. Chromatography
1. Let the solvent run to the top of the column bed and stop the flow. Add the sample in hexane and let it run to the top of the column bed. The flow rate should be 0.5 ml/minute or less. If the flow is too slow or about to stop disturb the packed sample on the head of column gently until adequate flow is obtained.
2. Rinse the sample tube with 2 ml hexane and transfer it to the column, washing down the sides. Let it run to the top of the column bed. Repeat this step 2 more times for a total of'6 rnL. Discard the hexane fraction. Rinse the column outlet with Elution Solvent.

Ополаскивают капилляр с образцом 2 мл гексана и переносят его в колонку, промывая боковые стороны. ?? Что-то не то..

Наносить вещество на колонку (жидкое или в растворе) следует по стенкам колонки...
